چکیده

strong correlation was found between the two variables (r = 0.86; p<0.0001), as suggested by linear regression analysis. The best results were obtained when the number of CD34 cells in PB was greater than 50 μL, while the worst results were related to a level of less than 10 μL (Figure 1). Only in 5 cases, because of the particular clinical history of the patients, did we perform leukapheresis when circulating CD34 cells were < 10 μL. None of these reached 0.5310/kg CD34 cells (mean value collected was 0.23, range 0.05-0.4310/kg). With more than 10/mL CD34 cells in PB we collected no less than 0.5310/kg, with rare exceptions. When CD34 cells exceeded 20/mL we usually reached more than 1310/kg. No statistical difference was found in patients with CD34 cell values ranging from 21 to 50 μL. Finally, in our hands, more than 50 μL circulating CD34 cells ensured a collection greater than 2310/kg in most patients. Though the quality of a leukapheresis does not depend only upon the absolute number of CD34 progenitors present, our data confirm that daily estimation of the circulating CD34 cell number by flow cytometry may guide our clinical decisions and may offer a useful tool for predicting the number of procedures to perform.

Apoptosis induction by retinoids in eosinophilic leukemia cells: implication of retinoic acid receptor-alpha signaling in all-trans-retinoic acid hypersensitivity.

Hypereosinophilic syndrome (HES) has recently been recognized as a clonal leukemic lesion, which is due to a specific oncogenic event that generates hyperactive platelet-derived growth factor receptor-alpha-derived tyrosine kinase fusion proteins. In the present work, the effect of retinoids on the leukemic hypereosinophilia-derived EoL-1 cell line and on primary HES-derived cells has been inve...

Regulation of myeloblastin messenger RNA expression in myeloid leukemia cells treated with all-trans retinoic acid.

Retinoic acid is known to induce differentiation of human myeloid leukemia cells in vitro. Recently, all-trans retinoic acid has been used to induce remissions in patients with acute promyelocytic leukemia, probably through differentiation of the leukemia cells. Myeloblastin (mbn) is a protease that has been identified in the human leukemia cell line HL-60. Downregulation of this protease can i...

Stat1 is induced and activated by all-trans retinoic acid in acute promyelocytic leukemia cells.

Treatment of freshly isolated acute promyelocytic leukemia (APL) cells and the myelogenous leukemia cell lines, NB4, HL-60, and U937, with all-trans retinoic acid (ATRA) results in a remarkable elevation in the amounts of Stat1 alpha and Stat2 proteins. Stat1 alpha protein levels are augmented by ATRA as a consequence of elevated amounts of the corresponding transcripts. The retinoid increases ...

Lineage-specific modulation of calcium pump expression during myeloid differentiation.

Calcium is accumulated from the cytosol into the endoplasmic reticulum by sarco-endoplasmic reticulum calcium transport ATPase (SERCA) enzymes. Because calcium stored in the endoplasmic reticulum is essential for cell growth, differentiation, calcium signaling, and apoptosis and because different SERCA enzymes possess distinct functional characteristics, in the present report we explored SERCA ...

Apoptosis induction with fludarabine on freshly isolated chronic myeloid leukemia cells.

BACKGROUND Fludarabine (FLU) is a fluorinated purine analogue with antineoplastic activity in lymphoproliferative malignancies. Recently, some in vitro reports have showed the effective role of FLU on the activation of apoptosis. MATERIALS AND METHODS The induction of programmed cell death (apoptosis) by fludarabine (FLU), an adenine nucleoside analogue, alpha-interferon (alpha-IFN), and FLU ...
